Can I set a custom field to be required?

Yes, for most types types of custom fields. To set a custom field to be required, select the "Is required" option in the popup dialog for the custom field. 

When a custom field is required, this means it cannot be empty. Although it will always start out being empty, once something has been entered, it is no longer allowed to be empty from that point on.
